Liver X Receptors (lxrs): Transcriptional Regulators of Macrophage Cholesterol Metabolism

چکیده

Liver X receptors (LXRα and LXRβ) are oxysterol-activated nuclear receptors. These sterolresponsive transcription factors regulate the expression of genes involved in intestinal cholesterol absorption and conversion of cholesterol to bile acids [1]. Since the macrophage plays an important role in host defense and immuno-inflammatory responses, particular interest has been paid to the role of LXRs in the control of macrophage gene expression and function. Altered macrophage functions contribute to the pathogenesis of many infectious, immunological and inflammatory disease processes, including atherosclerosis. Research reported over the last few years revealed important roles for LXRs in macrophage inflammation and cholesterol homeostasis with consequences in atherosclerosis development.

Decoding transcriptional programs regulated by PPARs and LXRs in the macrophage: effects on lipid homeostasis, inflammation, and atherosclerosis.

Macrophages play essential roles in immunity and homeostasis. As professional scavengers, macrophages phagocytose microbes and apoptotic and necrotic cells and take up modified lipoprotein particles. These functions require tightly regulated mechanisms for the processing and disposal of cellular lipids. Under pathological conditions, arterial wall macrophages become foam cells by accumulating l...

Transcriptional regulation of macrophage cholesterol trafficking by PPARalpha and LXR.

PPARs (peroxisome-proliferator-activated receptors) and LXRs (liver X receptors) are ligand-activated transcription factors that control lipid and glucose metabolism, as well as the inflammatory response. Regulation of macrophage function in inflammation and atherosclerosis.

Atherosclerosis can be considered as both a chronic inflammatory disease and a lipid metabolism disorder. Innate immunity pathways have long been suspected to contribute to the initiation and progression of atherosclerosis. This suggests that crosstalk between lipid metabolism and innate immunity pathways plays an important role for the development and/or the prevention of atherosclerosis. Howe...

Identification of macrophage liver X receptors as inhibitors of atherosclerosis.

Recent studies have identified the liver X receptors (LXR alpha and LXR beta) as important regulators of cholesterol metabolism and transport. LXRs control transcription of genes critical to a range of biological functions including regulation of high density lipoprotein cholesterol metabolism, hepatic cholesterol catabolism, and intestinal sterol absorption. Although LXR activity has been prop...
